High Throughput Screening Identifies CDK Inhibitors as Inflammasome-Activating Vaccine Adjuvants

چکیده
Abstract Several FDA-approved adjuvant formulations, such as saponins and lipid nanoparticles, have been shown to mediate their effects in part through the NLRP3 inflammasome. These formulations are among most potent adjuvants use, but they limited by poor synthetic control immunotoxic side effects. New small molecule targeting inflammasome would allow broader implementation of subunit- nucleotide-based vaccines against poorly immunogenic antigens. In this work, we leverage high throughput screening immunomodulatory compounds identify cyclin dependent kinase (CDK) inhibitors a novel class inflammasome-activating adjuvants. CDK anti-cancer drugs which block cell cycle progression prevent tumor proliferation. When co-administered with Toll-like receptor (TLR) agonist, observe that these induce robust NLRP3- Caspase-dependent IL-1β secretion primary murine human dendritic cells. formulated TLR4 agonist ovalbumin for use vaccination, were found enhance antibody production antigen-specific CD8 +T proliferation relative formulation controls. While greater work is needed formulate optimize adjuvants, understanding mechanism inhibitors’ adjuvanticity will both our expand case vaccination. Quantitative High-Throughput Screening Identifies 8-Hydroxyquinolines as Cell-Active Histone Demethylase Inhibitors
BACKGROUND Small molecule modulators of epigenetic processes are currently sought as basic probes for biochemical mechanisms, and as starting points for development of therapeutic agents. N(ε)-Methylation of lysine residues on histone tails is one of a number of post-translational modifications that together enable transcriptional regulation. Histone lysine demethylases antagonize the action of...
متن کاملInflammasome-activating nanoparticles as modular systems for optimizing vaccine efficacy.
Innate immune system activation is a critical step in the initiation of an effective adaptive immune response; therefore, activation of a class of innate pathogen receptors called pattern recognition receptors (PRR) is a central feature of many adjuvant systems. It has recently been shown that one member of an intracellular PRR, the NLRP3 inflammasome, is activated by a number of classical adju...
متن کاملHigh-throughput screening against thioredoxin glutathione reductase identifies novel inhibitors with potential therapeutic value for schistosomiasis
BACKGROUND Schistosomiasis, a parasitic disease also known as bilharzia and snail fever, is caused by different species of flatworms, such as Schistosoma mansoni (S. mansoni). Thioredoxin glutathione reductase (TGR) from S. mansoni (SmTGR) is a well-characterized drug target for schistosomiasis, yet no anti-SmTGR compounds have reached clinical trials, suggesting that therapeutic development ag...
متن کاملTrypanothione reductase high-throughput screening campaign identifies novel classes of inhibitors with antiparasitic activity.
High-throughput screening of 100,000 lead-like compounds led to the identification of nine novel chemical classes of trypanothione reductase (TR) inhibitors worthy of further investigation. Hits from five of these chemical classes have been developed further through different combinations of preliminary structure-activity relationship rate probing and assessment of antiparasitic activity, cytot...
متن کاملIdentification of QS-21 as an Inflammasome-activating Molecular Component of Saponin Adjuvants.
Many immunostimulants act as vaccine adjuvants via activation of the innate immune system, although in many cases it is unclear which specific molecules contribute to the stimulatory activity. QS-21 is a defined, highly purified, and soluble saponin adjuvant currently used in licensed and exploratory vaccines, including vaccines against malaria, cancer, and HIV-1. However, little is known about...
